Kytococcus sedentarius, is a Gram positive bacteria which degrades keratin making it responsible for the disease pitted keratolysis. K. sedentarius is also the only known producer of antibiotic monesin A and B. The genome of this organism has been sequenced and computer annotated. However manual annotation is required to confirm computerized annotation. The purpose of this study was to test the hypothesis that the genes, which encode for the enzymes involved in biosynthesis of methionine from L-aspartate have been correctly annotated. Different online tools were used for the manual annotation of these genes. The local sequence alignment was carried out using Basic Local Sequence Alignment Search Tool (BLAST). The structural analysis of each of the genes was carried out using TIGRFAM, PFAM and PDB databases. The enzymatic functions and their presence in different pathways was analyzed using KEGG, Metacyc. The gene sequence was checked for the presence of an alternate open reading frame using the IMG-ACT tool page. The gene was checked for evidences of it being a pseudogene and for the occurrence of a gene duplication event. The phylogenetic tree was constructed using a phylogeny.fr tool. All the genes were manually annotated and were found to be well conserved. The proposed annotations for the gene were accepted.
